Cynomolgus PBMC: An Indispensable Bridge for Zoonotic Disease Research

2026.04.08

Peripheral blood mononuclear cells (PBMC) from cynomolgus macaques serve as an irreplaceable bridge in zoonotic disease research. Benefiting from their genetic background, immune system structure and physiological functions that are highly similar to those of humans, cynomolgus PBMC have become a critical tool for dissecting pathogen-host interactions and assessing the safety of vaccines and therapeutic strategies.

In studies of highly pathogenic viruses such as Ebola, Marburg, Lassa fever and Crimean-Congo hemorrhagic fever, the cynomolgus macaque model precisely mimics human disease progression, and its PBMC samples provide direct evidence for the dynamic monitoring of immune responses—including T/B cell activation, cytokine profiling and antibody production. For example, in research on VSV-vectored vaccines against Marburg virus, analysis of transcriptomic and receptor repertoire changes in PBMC has identified key pathways underlying protective immunity; in Lassa fever mucosal exposure models, PBMC have helped reveal the association between interferon signaling and immune cell dysregulation.

In addition, in Mycobacterium tuberculosis infection studies, the combination of PBMC and MHC tetramer technology has successfully tracked the distribution of pathogen-specific T cells in lymphoid tissues and granulomas, providing targets for vaccine design. Cynomolgus macaque PBMC are also widely applied in cross-immunity evaluations against pathogens such as influenza, Zika virus and SARS-CoV-2, as well as in the efficacy and safety validation of monoclonal antibody drugs.

Notably, the high polymorphism of MHC and KIR genes poses challenges for individualized research; however, precise genotyping can optimize animal screening and improve experimental reproducibility. With the deep integration of single-cell sequencing, immune repertoire analysis and other technologies with cynomolgus macaque PBMC, this model will continue to drive the translation of zoonotic disease prevention and control strategies from the laboratory to the clinic, laying a solid scientific foundation for global public health security.
