In modern drug discovery, early-stage screening and preclinical evaluation are essential for determining whether a candidate compound is suitable for clinical trials. While animal studies have long played a critical role in pharmacokinetic and toxicological research, regulatory emphasis on ethics and the global shift toward the 3Rs—Replacement, Reduction, andRefinement—are accelerating the adoption of in-vitro systems that are more ethical, efficient, and cost-effective.
Among these in-vitro models, Liver Microsomes and Liver S9 Fractions remain indispensable tools for evaluating drug metabolism. They provide a controlled and reliable platform to study metabolic pathways, intrinsic clearance (CLint), enzyme activity, and potential drug–drug interactions.
Liver microsomes contain a broad spectrum of Phase I metabolic enzymes—most notably the cytochrome P450 (CYP450) family—which play a crucial role in the oxidative, reductive, and hydrolytic metabolism of xenobiotics. By simulating the liver’s primary metabolic processes, microsomes enable researchers to:
* Assess intrinsic clearance and metabolic stability
* Predict ADME characteristics (Absorption, Distribution, Metabolism, Excretion)
* Identify reactive or toxic metabolites

Figures 1 and 2 depict stability experiments conducted on Cynomolgus Monkey liver microsome products.

Figures 3 and 4 depict stability experiments conducted on Beagle Dog liver microsome products. Beagle dog microsomes show no significant change in clearance rate after two freeze–thaw cycles, confirming exceptional stability.
